spriting question

I never really looked into tutorials on spriting, but i recently did. Here's the video I watched:

Now i do all of my sprites in MS Paint as well, but in the video, the guy uses the eraser tool to recolor his sprite. When I first saw this, i reacted" is that even real?" because if it were, i could make sprites in a breeze. So i decided to try it out, open Paint, and started recoloring skuntank with purugly. I do what the video says, but when I right click the eraser tool, nothing happens. Can anyone tell me how he did that? Because i use the bucket-fill tool for recoloring, which is very tedious.
Also, in what other programs do you do your sprites?
 
You right click the colour you want to change the other colour into, and you left click that other colour. Then you click eraser the tool, right click, and hover over the colour you want to change.
 
if you are following the video exactly and it still doesnt work its probably because the sprite has a background. I dont relally understand this but i understand how to get around the problem.
What you do is click the Select tool and then pick the second type in the box below now draw aound your sprite move it and put it back. now the eraser tool will work for colour replacement. My only problem is i dont understand how to save them without the white background in MS Paint

It took awhile for me to understand you, but it totally works! Thanks so much. Without the white background.. Do you mean transparency?
 
Well, there are quite a few spriting tutorials on YouTube, I you know where to look.
